Immature pods, ranging in age from 15 to 30 days after pollination, were collected from greenhouse-grown plants. At harvest, seeds were removed from pods and isolated pod walls were collected and immediately frozen in liquid nitrogen. Pod walls were pooled for mRNA extraction. |

| Description |
cDNA was prepared from polyA+ enriched RNA. The cDNA was directionally ligated into the Uni-Zap XR vector from Stratagene and packaged using the Gigapack III Gold packaging extracts. Plasmids containing inserts were excised from the recombinant lambda-Zap phage using ExAssist helper phage and propagated in XLOLR cells. |
