cDNA was prepared from polyA+ enriched RNA from roots harvested at 10, 17, 22, 31 and 38 days post-inoculation with Glomus versiforme. The cDNA was directionally ligated into the Uni-Zap XR vector from Stratagene and packaged using the Gigapack III Gold packaging extracts. Plasmids containing inserts were excised from the recombinant lambda-Zap phage using ExAssist helper phage and propagated in XLOLR cells |
